Mechanism of action
Fluconazole inhibits the enzyme lanosterol 14α demethylase, which is dependent on cytochrome P450. The main function of this enzyme is to support fungal cell synthesis by conversion of lanosterol to ergosterol. Fluconazole has a free nitrogen atom on its azole ring that binds with a single iron atom on the heme group of lanosterol 14α demethylase.
This process inhibits ergosterol biosynthesis and arrests further fungal growth and destructs the plasma cell membrane of fungi.
